Malgrado il tempo a disposizione fosse pochissimo, un mese circa, Donizetti accettò l'incarico, insistendo che la composizione del libretto fosse affidata a Felice Romani, forse il librettista più famoso di quei tempi. L'opera ebbe immediatamente un enorme successo e fu replicata per ben trentatrè volte. Felice Romani, che aveva già scritto il libretto per Anna Bolena, portò a termine il suo incarico in meno di una settimana, basandosi su quello che Eugène Scribe scrisse per Le Philtre, un' opera che Daniel Auber compose nel 1831. Nonostante le origini francesi, il testo proponeva dei personaggi riconducibili all'opera buffa italiana del diciottesimo secolo: il dottore-ciarlatano che viaggiava di paese in paese per vendere improbabili intrugli (Dulcamara), il giovane e gagliardo ufficiale (Belcore), il ragazzo di campagna semplice ed ingenuo, (Nemorino), la fanciulla volitiva dal cuore tenero (Adina). Grazie all'eccellente libretto di Romani ed alla musica di Donizetti questi personaggi superano le stilizzazioni del dramma buffo ed acquistano spessore e profondità psicologica. L'Elisir d'amore non può essere definito un'opera buffa nel senso stretto del termine. E' piuttosto una commedia brillante con molti punti in comune con le opere semiserie, a partire dal contesto agreste. La scelta di questo tema è stata probabilmente una conseguenza dei successi tributati all'epoca a La Sonnambula di Vincenzo Bellini....
